In silico prediction of the mechanism of radix paeoniae rubra against menstruation disorder

Menstrual disorders are physiological conditions that affect women of all ages worldwide. To aid in the treatment of menstrual disorders, the effect of the herb Radix Paeoniae Rubra (RPR) was exam-ined for the synthesis of natural drugs. RPR is a traditional Chinese medicine employed for its effective treatment against menstrual disorders, but its underlying mechanism of action stands to be still uncertain. The aim of the current study was to determine the mode of action from a pharmacological standpoint of RPR in the treatment of menstrual disorders. The ingredients-target database was developed for RPR, which was followed by the elucidation of targets pertaining to menstrual disorders. The interaction of these targets with each other was investigated with the aid of PPIN which was constructed by STITCH and gene ontology (GO) enrichment analysis via its plugin ClueGO. From the ensuing literature review, 46 components in RPR were identified, from which 101 targets were accessed. These targets were screened which elucidated 8 potentially effective targets found to be affiliated with menstrual disorders. 5 GO terms were associated with the annotation clusters and abundant value of these potential targets, where majority of these terms were found to be associated with menstrual disorders. In the PPIN, the main hubs were found to be PTGS2, ALOX5, CYP1A2, and GSTP1, respectively. The conclusive findings of the study highlight the effective therapeutic role of RPR, in the management of menstrual disorders. However, the molecular mechanisms which are affiliated with the mode of action could only be established with the help of further in silico studies such as docking and simulation. © 2021, Colegio de Farmaceuticos de la Provincia de Buenos Aires.
