Abstract

Colon-targeted delivery of drugs is an active field of research focused on treating diseases affecting the colon, including inflammatory bowel diseases, irritable bowel disease, chronic pancreatitis, cystic fibrosis, and colorectal cancer. This approach holds significant promise as it enhances the effectiveness of therapeutics through localized treatment and reduces systemic toxicity. In recent years, nanotechnology has played a significant role in drug delivery by improving therapeutic outcomes. Among various nanocarriers, solid lipid nanoparticles have emerged as highly promising vehicles for colon-targeted drug delivery. This is attributed to their capacity to accommodate both lipophilic and hydrophilic drugs. In addition, SLNs provide a controlled drug release, site-specific targeting, good stability, and ease of manufacturing. The chapter explores the properties of SLNs that make them suitable for colon targeting, including lipid selection and surface modification strategies. Fabrication techniques for colon-targeted SLNs and methods of characterization are also discussed. Furthermore, this chapter highlights therapeutic applications of colon-targeted SLNs, including drug delivery for colon cancer, treatment of inflammatory bowel disease and the delivery of probiotics and nutraceuticals. This chapter also sheds light on the challenges and future perspectives of colon-targeted SLNs.
