Abstract

The interplay between the gut microbiome (GM) and environmental toxicants (ETs) is of paramount importance in shaping human health, offering both avenues for exploration and hurdles for intervention in future research. This chapter consolidates existing knowledge to delve into the intricate interactions between environmental chemicals and the GM, highlighting their profound influence on metabolic pathways, epithelial barrier integrity, and host-microbial dynamics. Through a review of recent studies, we illuminate how prevalent ETs such as arsenic, trichloroethylene, polycyclic aromatic hydrocarbons, and organophosphate insecticides disrupt the GM's composition and function, thereby impacting host health through various physiological mechanisms. Moreover, we underscore the pivotal role of the GM in directly metabolizing ETs, thereby influencing their toxicity and potentially exacerbating detrimental effects. The perturbation of the GM epithelial barrier by ETs underscores the intricate interplay between the GM and host tissues, rendering individuals more susceptible to toxicant-induced harm. The integration of multi-omics methodologies provides a holistic comprehension of the GM's involvement in the metabolism of ETs and subsequent host responses. By leveraging diverse biological datasets encompassing DNA, RNA, proteins, and metabolites, researchers can unravel complex interactions and develop personalized therapeutic approaches and predictive models to mitigate the adverse impacts of environmental exposures on human wellness. This chapter aims to propel our understanding of the biological pathways orchestrated by the GM following exposure to ETs by addressing current challenges and proposing innovative strategies. Ultimately, this research contributes to the development of targeted interventions aimed at preserving the composition and functionality of the GM, thereby fostering human health and well-being amidst environmental adversities.
